Take a look at the documentation James Rich has put together:

Specifically, look at the section regarding the ".tn5250rc" file (page 5).

Here's a snippet of what mine looks like:

To start my "A" session I have an icon on my desktop that executes
"xt5250 PGBENNYA" and will connect to the host I have defined in my
.tn5250rc file.
