Alt-W, P, then in the filter box 'parsers' (without the quotes) is the
fastest way to access the preference. I don't think there's a way to
assign an action to modify a preference.

Making a change to that preference touches this file:
...\.metadata\.plugins\org.eclipse.core.runtime\.settings\com.ibm.etools.iseries.edit.prefs

Within that file:
com.ibm.etools.iseries.edit.preferences.parser.signature.enable=true
com.ibm.etools.iseries.edit.preferences.parser.signature=\!rdi\!

I tried changing this file outside of the preferences dialogue, but RDi
did not recognise this. My best guess is that RDi needs to 'reload' the
preferences.
