What specific error message are you getting?  What version of Access are you
using?  Which ODBC?

I am unclear on where the error is popping up.  Are you getting an AS/400
signon window?  Are you seeing a list of file names available from your
library list to be imported?

We were getting an error where we would select the AS/400 file and click
Import; after about 5-10 seconds, an error window would appear, something
about a "file unavailable" or somesuch, even though the file was not at all
allocated on the AS/400.  Our fix was to reinstall Access.

I highly doubt that the drive letter issue is a problem.
