I'm not using MMail, but we experienced a similar issue some years ago when we started sending email from the iSeries using SNDDST. The issue turned out to be a routing table problem on the iSeries where we had specified the wrong gateway. In essence, what we found out is that the message would bounce around the network repeatedly until it found the right address. Take a look at your routing tables to see if everything is correct there. Also, try TRCTCPRTE to see the path it's using.
