Forget twinax and the PC.  IMO The easiest, simplest method is to use a
network printer adapter such as a JetDirect or Marknet.  Using a wireless
network adapter could be a decent option too.  The PC might be an option if
you also need to print from PC software such as Label Matrix but going
straight from the AS/400 to the printer tends to simplify things, requires
less maintenance, and eliminates potential cabling headaches.

We use Datamax printers here. Mostly Prodigy printers that we bought way
back in the early 90's. We've got about 30 of these printers and they run
ALL the time. By that, I mean from the start of first shift to the end of
2nd shift (approx 16 hours) they are printing continuously, as the workers
pull one label out of them at a time and place them on the packages.

We've also had some experience with the newer Datamax printers (such as
the I-4208) and if I had to start over again,

We use TL Ashford's Barcode400 software (http://www.barcode400.com) to
generate labels, though you can also write your own programs that generate
them if you're willing to program in all of the printer escape codes
(which I've also done, it's not difficult... the only trick is that there
are some control characters that you have to figure out how they'll get
translated when sent to the PC to end up with the right results. WSCST
helps a lot.)

You can definitely do it without a PC.  There are adapters available that
even allow you to attach the printers via Twinax. :)
