Large hard drive partitions, yes, at least from DOS 5.0 on. Maybe not the
huge ones you see on WinDoze boxes, but the DOS side of my cheap generic
box has a bunch of big ones.

Graphics, YES! GEM and GEOS are only the two best known non-WinDoze GUIs
that will run under DOS.

USB and Firewire, well, I haven't been looking very hard, as I don't
really need them on the DOS side. On the other hand, PC-DOS 2000 does come
with PCMCIA support (which I use fairly extensively on my notebooks).
