At "one time", IBM had a special lab set up for researching and dealing with very large files on the 400. I have not read or seen anything about that in a while, but I do recall a story on the lab in one of the trade rags. If it still exists, it would almost certainly be worth your while to contact those folks.
That's a pretty large file for almost any system....
