Mike, You should be able to go directly from any 6.x release to 7.0.1 (definitely to 7.0, so you should be able to do 7.0.1 as well). I would definitely get up to 7.0.1 before upgrading to V5R4 so you don't have to worry about whether the Domino or OS upgrade caused a potential issue.
