CHKOBJ OBJ(&LIB/&OVLNAME) OBJTYPE(*OVL)

And, you can call the same command from RPG via QCMDEXC (or system(), or
QCAPCMD, etc)

You could also use the "retrieve object description (QUSROBJD) API" and
check to see if it finds the object successfully..  but that takes more
code, and it's probably not necessary for something this simple.
