I have two boxes next to each other, one XP, one Vista Business. The XP machine
can map fine, but Vista can not, it would be nice if it was that simple, but we
are unable to map to our v5r1 box, or our v5r3 machine.

I'd be willing to bet that your running OS v5r4 on your system and are able to
map it as a result. IBM "officially" supported vista in v5r4 but made no
promises on previous versions so its been quite a challenge so far to figure
out how to get this to work, from disabling the LM hashing, to mixed mode
passwords, to SMB digital signing. Nothing works so far.

It is the same as in XP. In Windows Explorer do Tools - Map network drive.
Select the drive letter and enter \\iseries_name\shared_folder in the Folder
field. Check Reconnect at logon and select 'different user name' if you don't
sync Windows & iSeries ID/passwords.

I just did in from my Vista Ultimate laptop over VPN to one of our machines.

Alternately, in Explorer you can enter \\iseries_name\root in the address bar,
enter credentials if prompted, and let it populate the list. Once the icons
show as network folders (they have a little network pipe overlaying the
folder), you can right-click and Map network drive from there.
