For a graphic approach:
iSeries Navigator/Operations Navigator
Expand the listing for the selected iSeries 
  Network
    Configuraton 
      TCP/IP Configuration
          Right click on Routes, choose New Route, and a wizard will pop up
to prompt for details of the route definition
