I assume you are pinging by IP address.

What kind of Sub net mask is the VPN assigning? Could the AS/400's IP
addresses be out side of that sub net?  You could try a trace route
(TRACERT) and see where it goes/stops.

To ping form the AS400 from a command line key in
ping 'the.ip.address'
you need the ' around the ip address.
