David, 
Your AS/400 is probably not using DNS or your DNS server does not have
an MX record pointing to the Exchange server as the mail server. Check
your AS/400 hosts table and ensure that the host name entered for the
mail server corresponds exactly to the domain name used on the email.
Alternatively specify a mail router on the SMTP set up.
