Well, assuming that the bytes= is not zero then you are able to make a connection from your PC to the 400. PING does not verify that TCP is active on the 400, only that the interface (IOP) is active. Did you check whether TCP is started and also whether the TELNET server is started on the 400? Does the TN5250 application give you any useful error messages or just the usual windoze crap -- "Error: It didn't work. OK?" Proxies can be used by all TCP/IP stacks but SOCKS requires TCP clients that understand a SOCKS server. Good implementations of TCP (such as the one that comes free with OS/2) provide SOCKS-aware clients. I believe WinXX requires third party clients.
