If I do specify Kerberos as mentioned below, the spool file filter stops working.

Some nice guy at IBM pointed me to the "Remote System -> IBM i -> Authentication" preference page, where you can select "Kerberos" for "Authentication options". I assume that this page has been added with RDi 9.5.

There is nothing more that I can do to make iSphere supports Kerberos, since iSphere completely relies on class IBMiConnection, which is part of RDi.

Honestly I do not know what I should have to do, to make iSphere supporting Kerberos.

iSphere utilizes IBMiConnection and AS400, which are classes shipped with RDi/WDSC.

Hence I assumed, that iSphere should be able to support Kerberos, as long as Kerberos is supported by "com.ibm.etools.iseries.subsystems.qsys.api.IBMiConnection" and "com.ibm.as400.access.AS400".
