If it's accessible from the internet you may want to try
http://www.zmailer.org/mxverify-cgi.html

You could try these steps from a Windows DOS command prompt
telnet myibmi 21
This tells you to telnet into your IBM i on port 21 (email)
Enter in this command
RSET
It should echo back
250 flushed
Then enter this command
MAIL FROM:<postmaster@xxxxxxxxxxxxx>
It should echo back
250 OK
Then enter this command
RCPT TO:<postmaster@xxxxxxxxx>
It should echo back
250 OK
Now you know you're connecting pretty good.
At this point I normally enter
QUIT
