Once configured, the POP3 email on the iSeries will only work
if the emails are forwarded from Exchange.

Well, the IBM i POP server "can" receive email from anywhere in the world if you set up the routing that way, but I think Ken's suggestion about using a filter in Exchange to forward to the IBM i server (presumably on the internal LAN) is a good idea. Otherwise your IBM i POP server would end up handling a lot of SPAM.

Regarding the sender email account used by the SNDDST command, use CFGTCPSMTP option 1 or 2 to display the email address associated with your IBM i user profile. I gather it points to an account on your Exchange server.
