Let's back up a little. What problem are you trying to solve?
Are you wanting to send mail from the as/400 using SNDDST (and like
utilities) while still send/receive from Domino from/to the Internet?

For both SMTP on the AS400 and on the Notes 6.0 server, what settings are
needed in NOTES and on the SMTP server for them to share the same port 25?
