This should work:

Have the print file opened under user control
For each different print queue
  Issue an override to the print file via qcmdexc to point it at your
desired output queue
  Open the print file
  Print everything for that output queue
  Close the print file

You could do this for every PO but it would be more efficient to sort the
input by print queue if possible.  Be careful with the override to be sure
it stays in scope.

I have created an RPG/400 program that generates our Purchase Orders.

Depending on the state of the supplier the output needs to end up on any of
three printers.

In any given purchase order run we will generate purchase orders for all
three printers.

I am happy to generate one spool file per purchase order (rather than having
all the PO in one large spool file)

Question:
How can I control the output queue of my spool file(s) from within my
program.
