According to a consultant that we have used in the past (and who really
knows his stuff), you can migrate directly from OS 4.4 to 5.1 without any
issues.

One problem (or opportunity) that you may run into is that your security
profiles, user queues and physical files will not migrate to OS/400 5.1
unless you are already at OS/400 version 4.5 or higher. 

When we upgraded to 5.1, we first had to go from 4.3 to 4.5 and then to 5.1
in order for us to be able to use our existing security and physical file
structure.

What version of the OS are you running now?
