One way is to FTP the file over. If the file sits in a specific folder on the network then a CL program can go out and retirieve it and bring it in to the iSeries using FTP.

Another way is to set up an ODBC connection, there are many tools that can move data either way over ODBC such as SQL server or maybe even Micrsoft Access. The ODBC drivers come free with client access then you can use any ODBC compliant application.

I would recommend saving the file as tab delimited. Comma delimited text files will get confused if any of the text values conatin commas.
