The file name for the System-wide SMTP Alias File is (if the brain is
awake) QATMSMTPA.  It's fully documented in the TCP/IP manual.  We
routinely add new Internet Email addresses to the system by using the
ADDDIRE (Add Directory Entry) command, then writing a record to the Alias
file.

If a user has already sent -you- a message, there's nothing to do (as long
as you have automatic enrollment turned on):  the incoming mail has already
added a directory entry and an alias to the alias file (if needed; see the
TCP/IP manual for times when you get -only- a directory entry).  Just reply
to the incoming mail, and let the system sort out the pathway back to the
originator.
